I am just speculating here since I never cared for Jailbreaking my Kindle. The recent *lighter font* messup released by Amazon caught me with the idea of Jailbreaking the Kindle as soon as a new JB is available. Only to avoid future *surprises* by Amazon.
Short answer: No

Longer answer:
The update_*.bin packages are cryptographiclly signed.
The firmware's installer tests the signature with the signature certificates in the Kindle's key store.

Our (mis-named) "Jail break" adds the MR developer's certificate to the Kindle's key store.

The tool to take apart (and put back together) the update_*.bin packages is named: KindleTool
The most recent version is always to be found in NiLuJe's snapshots thread top post.
