book64 - I have a several physical libraries ranging from a 400 to almost 70,000 books, my start time is about 6 or 12 secs* no matter which library (or virtual library) is opened at the start.
Composite custom columns (columns created from other columns) can slow things down; the extent depends on what is in the template code used to create them. Starting with an empty library would give indication if that's a cause.
It could be an over aggressive AV. Try disabling it, if that has any effect, try giving calibre dedicated temporary and configuration directories via the relevant
calibre environment variables and exclude those folders from your AV.
* Variation is due to the data drive going to sleep when idle - my choice.
