It shouldn't and it doesn't here. But, it will if you change the metadata that is used in the save template. I do this all the time. I fix spelling and layout errors, put the book in the send-on-connect list and the book is replaced the next time I connect.

I should add that resending a book that is already on a Kobo device only works properly if there are no structure changes. The books aren't reprocessed by the device, so any changes to the ToC or the internal file structure can cause problems. Plus, the metadata is not reread. If the changes are spelling, CSS or formatting, it works.
