You need to use the full path to calibre-debug

/Applications/calibre.app/Contents/console.app/Contents/MacOS/calibre-debug

Although I have to say as far as remember calibre-debug has no -R option, so I'm not sure what you are trying to do. If you want to run a plugin, the option is lowercase -r
