If you run a conversion, then there will always be differences between the input and output.

Have you set the tablet profile yet? Has that solved the problem of image quality loss? Do not forget that when reconvert a book after changing the global settings then you need to tell Calibre not to use the stored values for that book if you want to revert to using the current global settings.
