I don't know! Interesting question. My workflow is a bit different, in that I upload the epub to the KDP and let it convert there, before downloading the "mobi" and previewing it.

Starting fairly early on, when I uploaded html files, I found that ANY image at the front of the book would be suppressed in favor of the cover, even if it was a legitimate frontispiece and no cover at all. So I learned to move illustrations past the title page.

For at least a year, I uploaded epubs with cover included, and they worked just fine. Then someone pointed out to me (I think in January 2012) that a newly published book had TWO covers when downloaded. I bought a copy, and indeed it did. Since that time I've not added a cover until I go to build the version intended for other retailers.
