@psychobunni - you could use Windows Disk Manager to put it back to E:

This is one reason why it's not a good idea to put non-portable (installed) software on removable drives. Installed software is software that the Trusted Installer knows about, and that shows up Control Panel->Programs and Features. The location of installed software is recorded in the Windows registry, so by changing the drive letter you created a mismatch, and that's what causing the problem.

If you use the portable version of calibre you wont run into this issue.

BTW this is not a calibre issue as such - you control where software is installed, and Windows does the installing/upgrading - not calibre itself. The Welcome Wizard is about calibre library locations and ebook-reader devices.
