Once a book is on the Kindle, the Kindle itself generates a folder called whatever.sdr. This folder is NOT created by the Calibre plugin and probably won't be in your Calibre library at all.

So you need to go to wherever you generated the .asc file, copy the file from there, then look at the filesystem on your Kindle, find the .sdr folder, and paste the file.
