If Windows I think all you need is a Windows shortcut that runs calibre but sets the "Start in" directory to the directory that contains the translation.

In my case I tried
  • Right click on desktop and selected New Shortcut
  • Typed in C:\Program Files\Calibre2\calibre.exe as the item to make a shortcut to
  • Typed in Test Translation as the name of the shortcut
  • Once the shortcut was made, located it on my desktop, right clicked and selected Properties
  • Changed the Start In field to the directory that contained my new PO file
  • Selected the OK button

Now I have a new shortcut for calibre that launches it from the directory containing the translation.
