Your Nook book files will be in username/Documents/My Barnes & Noble Ebooks/emailaddress/ on your Mac. I should probably remember to put a link to it in my Finder sidebar because I'm lazy (and now that I've done that, it's shift-command-T to add stuff to the sidebar). That makes it easier to point calibre at the right folder for adding books.

If you've been downloading your books and stripping DRM all along, you just need to use the Nook app to download any new books. If you haven't, you'll probably need to argue with the software to get them all downloaded and added to your calibre library. Once the DRM is stripped, you can read them whereever you like. You do NOT need to re-download books where you've already stripped the DRM.
