This tripped me up at first as well, and as far as I can tell there's no documentation to help you. It turns out to be a simple problem though...
Right-click on your HotSync icon, select Custom, select JPluck, select Change, enter the JXL to use and the default conversion profile, change the destination if you like, and make sure the conduit is enabled.
Now everything should work.
