Seems strange that one way works, which I'm happy with but deep sadness not the other.

If I understand you correctly, the wireless device driver is connecting? After you get "Syncing with Calibre" you have "Disconnect" in the action bar? If so, then things are working as expected. Some explanation might help.
When you connect to the content server, CC is a "pull" client. That means that you see in CC the books available on the server and can choose which books to download (pull) to CC. However, when you connect as a wireless device, CC is a "push" client. You use calibre itself to send (push) books to CC. After you connect, look at calibre. You will see a new toolbar icon at the top, Send to Device. If you select a book (or lots of books) in calibre and push that button, those books will be sent to CC.
Another benefit of connecting as a wireless device is that CC asks calibre for any new metadata (book information). If in calibre you change any information about a book (title, tags, series, whatever) after you have sent it to CC, those changes will be sent to CC the next time you connect as a wireless device.
