Environment variables are set as per the second link, in the OS. Any running program will inherit all environment variables. calibre will use the value set in that environment variable if available.

If it was to be set from within calibre, then a) it would have to go in the config directory. catch-22 -- which one? and b) the manual would not tell you how to set a system environment variable.
