I read medical textbooks on the iPad using the Goodreader app. My 0.2 cents -> Forget e-ink for medical textbooks. Too slow to read, too slow to annotate, and only monochrome. I will also have to disagree with the suggestion of an 8 inch tablet; it's too small for medical textbooks. The iPad with it's 10 inch screen is barely sufficient, and I'd jump all over the rumoured 13 incher if they did release it.
