Are you using an account with administrative privileges? Windows should prompt to raise the privileges if you aren't, but something might be going wrong with that.

Cyphox's suggestions are good. Plus I would try another cable. And another PC. You can also try USBDeview to make sure the Kobo drivers are completely uninstalled. Find any reference to Kobo devices in the list, select and uninstall them. There will probably be a lot of drivers for other devices. Removing any that are no longer used might help. After that, reboot and see what happens.
