The <strong> or <em> properties are declared in epub html code. An epub reader like Moon+ should interpret it and find the proper font to display it (rather than make fake bold and fake italic, or, slanted font). For example, if I put SegoeUI, SegoeUIBold and SegoeUIitalic.ttf in the same folder, Moon+ should use them all to display something like:

What's more, I hope that Moon+ will support advanced Opentype features like smallcaps and contextual ligatures, which are all the work done by font designers to make better reading experience.
