I'm not an OSX user so I dont have much idea what the problem on your system is, but usually you can find diagnostic output in console.app. You can also ask calibre to install its command line tools via Preferences->Miscellaneous. One of those tools will be /usr/bin/calibre which is a somewhat easier way to launch calibre than from inside calibre.app.
