Thanks for the advice, which I believe I'd anticipated. Just to be sure, I turned both Nook and PC off again, and tried sending again, but got the same error message. I'm wondering if it has anything to do with the latest calibre upgrade (1.6.0) as the problem started not immediately, but soon after I'd applied it.
