Or, your User Account Control (UAC) settings are too low to install software. If this is the case two routes exist:
If someone else is the Admin: You will need to speak to your Administrator (whoever that is) in order to have them update calibre. I would suggest letting the Admin know that calibre is updated every Friday, so that they're aware this is going to be a recurring issue.
If there is no one else as Admin: You might need to right click on Calibre's icon, Properties>Compatibility tab>Privilege Level>Run as Administrator>Apply>Okay.
