I assume you mean the cover displayed on the sleep screen, since you note that you have it set to display full screen. There are two ways to get the covers to always display full size:
1. Use Calibre to transfer the book to the device, with the "Upload covers for books" option set (and possibly the "Keep cover aspect ratio", depending on your preference).
or,
2. Edit the book so that the cover is displayed using SVG graphics, with the viewport set to the same size as the image. Most books seem to have been created with the assumption that the screen size is about 600x800, and need some fixing up to look right on larger screens.
