When you get the error what do you see if you use a file manager to examine the mounted USB drive(s).

I have a long-standing problem where the USB connection is lost when Calibre tries to synch with the Glo - the drive is still showing as mounted under Windows but is empty (Volume type: unknown, Staus : unreadable) and wonder if your problem is the same as mine.
