As to which series to start with, I read Alpha & Omega books first (not by design, it just happened that way). I think you can start with either one and it won't matter. There is a little overlap between the two series but nothing spoiler-y.
I've known a few people who felt very confused when they read the first Alpha & Omega book without reading the prequel novel in the anthology "On the Prowl". I would say it isn't necessary, but it does help you understand the backstory of how the hero & heroine meet.
