The file you download from Amazon is an azw, not a mobi. The two formats are related but the azw has DRM, which means it can't be opened by another Kindle or on the PC without removing that DRM. Removing the DRM is possible though using Calibre - you just have to have the original Device ID for which the file was created. I usually download the azw to my PC from Amazon, the run it through Calibre, but I suppose you could just drag the azw from the device itself and that would work the same way.
Note that Calibre itself does not remove DRM. There are third party plug-ins that do the removal.
