This is possible right now, it's called a custom column. In those you can put what you want. Here's how you can set up a "Read?" column:

Right click on any column header, and select "Add your own columns" at the bottom. Click the green + sign at the right, and enter read, Read? and select Yes/No as indicated in the left screenshot. Restart Calibre (Ctrl+R).

If you want to get fancy you can also make a column with multiple choices. After clicking the green + sign, enter the values shown in the right screenshot, enter a comma-separated list of whichever options you want. Restart Calibre (Ctrl+R).

On some readers you can also use such a column to create collections or shelves on your reader, I find this *very* handy for managing my reading queue.
