Just Install Calibre as normal:
1a) When asked about the Library Location, point it to the folder on the Memory Stick
or
1b) Make a New Location (on the hard drive), then Quit. Now copy the entire (USB stick)Library folder CONTENTS, into the new Library folder: replacing Metadata.db

IMHO 1b has great a speed (performance) advantage over 1a
