If you have the font correctly embedded in the EPUB, then the nooks will support the same font types (OTF and TTF) as ADE 1.8. On the nook, you will want to make make sure you have Publisher Defaults selected in the Text Options dialog, so that you are seeing your font instead of one of the default ones.

As for the font itself, I can't provide any concrete recommendations as to fonts, unfortunately I'm Tamil illiterate, so fonts covering Tamil are not something I've ever looked for.
