I don't own a Kindle, so this is an uneducated guess, but it sounds to me as though what they are doing is very similar to hibernating a laptop i.e. copying the entire contents of memory to a swapfile on disk and then powering down. The start up is just copying the swapfile back to memory, so no boot operations and it's much faster.
You don't get a boot sequence when you power up a hibernated laptop but once hibernated it is most definitely off. I'm sure I've removed/swapped batteries on hibernated laptops with no ill effect. If Amazon call this option "Off" then that's what it might be.
