According to Kovid Goyal, this would require a change in the source code.

Depending on the languages you're interested in, you have 3 possibilities:

1. Convert your book to the .mobi format and:
a) read it with Mobipocket Reader and a free Mobipocket dictionary
b) read it with Kindle for PC and one of the 7 free monolingual dictionaries

2. If it's a free ePub book, read it with the Kobo desktop application which comes with free bilingual dictionaries for English, French, Italian, German and Spanish and a couple of Public Domain books in several languages.
