As far as I know, all the Calibre font filter does is remove any existing font-family assignments in the epub's existing css. This should allow the PRST1's select-a-different-font standard feature to actually change to the font you have selected (rather than do nothing). I don't believe it will fix the 'unpredictable bold and/or italics' problem.
