In the previous post you mentioned you wanted to edit your collections manually, but this post indicates you want to manage them using calibre. Editing manually means using the Edit Collections option of the plugin to show a GUI that allows you to edit the collection names directly on the Kindle without using any information from calibre. I'm guessing you really do want to use calibre to manage your Kindle collections, but just want to enter specific collection names and not create them based on author or series, etc.

So if you've already imported your collections from the kindle to calibre and made changes to the column, what you need to do now is use the Customize Plugin option to select 'Create' next to your column name so that the plugin knows which column to use. Then you run the Preview Collections to show a report of what collections will be created. If it looks ok, you can run Create Collections to save them to your Kindle. Then you just need to Restart your Kindle so that it loads the information (Restart is not just power off/on - use menu->settings, menu->Restart).

[EDIT] If you have any books on your Kindle that are not in Calibre, then you will need to manually create separate collections for them on your Kindle. Or import them into calibre and assign them to a collection.
