You missed off the semi-colons.

Your text should read: This is a test text using »guillemets«.
(Conversion test and result attached).

Converting this to ePub in calibre does result in the correct symbols.

You can also edit the ePub directly in calibre after conversion, (thus preserving your original text file), using the Tweak ePub option, from where you could do a global find/replace of your incorrect symbols.
