Depends on what track you want to start with:

Anhk-Morpork Watch - Guards, Guards
Witches of Lancre - Wyrd Sisters
Death - Mort (Tho I also find Hogfather easy to get into as well)
Rincewind - Interesting Times (tho, admittidly, I haven't read a lot of those books.)
Moist von Lipwig - Going Postal

Then there are the Tiffany Aching books, which are mostly their own thing and should be read in order ("Wee Free Men", being the first book.)

Personally, I find the Watch books are the best ones, IMO, but tastes vary.
