I have only about 250-300 books on my 350. Today I got my new 350 that I got as my spare (since I hate the T1), and did a comparison.

From sleep mode to open to my current book being read, the time between the brand new with nothing on it and my 10 month old with the 250-300 books on it was identical.

Powered fully off though and then restarting both, my older 350 with the books on it did take about 12 seconds longer. The progress meter "loading book" on the first page progresses along at the same pace and they reach the home page at the same time, but at that point though is where the extra time comes in for the reader with the books on it. The circular wait icon comes up in the middle of the home page while it's loading everything.

But as others have already said, you should always put it in sleep mode after reading, not fully turn it off nor let it sit until it powers itself off. The battery will last several weeks and you never have that wait time to fully restart it.
