I would definitely download as many applications as you could for testing, but unfortunately most of the desktop applications use different rendering engines than their hardware (i.e. the Nook app verses the Nook hardware) so they aren't a very good way to test for devices. The only real way to test for devices is to test on those devices.
The book view in Sigil is not a very accurate way to test how a book will display (it's much more forgiving of CSS errors that ADE is).
With large images I try not to exceed 600 pixels wide by 730 pixels high (the actual usable screen resolution of my nook).
