There weren't any changes that could affect the database in 0.8.12. Run calibre ina command prompt as

calibre-debug -g

and post the detailed error (if any) you get there.

Also try a clean install, i.e. uninstall, delete c:\program files\calibre2 and then re-install. Doing so will not affect your books/settings.
