Really the main thing is the overall touch experience - especially how quick and easy it is to look up words in the dictionary (my biggest pet-peeve on the original). I'm amazed at some of the BN stores out there - you can tell the Fargo store employs a ton of booklovers and readers because they are all over the new Nook as well. They had just gotten to the two display units and I had to pry it out of the hands of one of the employees...

I'm worried though - they are expecting their shipment today and they're not sure they're going to have enough to fill all of the pre-orders... (as you can tell, being blizzarded-in five months of the year, we have a lot of readers in this area).

Went to B&N yesterday and played for almost 30 min with one. Agree with almost everything. Fantastic piece of hardware and feels great! Great ergonomics and feels solid. But I did not find the dictionary lookup to be faster than Kindle or really fast. It is a two step action on the new Nook. On Kindle, you get a word's meaning, at least a short one, immediately after you position the cursor close to the word. But certainly can be more comfortable for some people.

The screen is also sharp and crips, same as Kindle 3. A big improvement if you had a Nook 1st generation, you will notice it at 1st eye.

For people looking for a new eink reader and if they don't read PDFs or technical books, I would recommend the new Nook without a blink of an eye. I think is even more value that the Sony, which is also touchscreen.
