Yes. Here is what you need to do:

You must eject, not just "safely remove" the Nook. With the Nook plugged into the USB cable, you'll see an icon in the system tray (lower right corner of the screen) that lets you "safely remove" the device. Don't use that. Instead, open "My Computer" or "Computer", depending on your Windows version. There, you'll see a list of drives--your hard disk will be C:, probably a CD or DVD drive, and so forth. One of those will be for the Nook. Right-click on that and select "Eject" from the context menu. Now you'll be able to use the Nook while it's still plugged in.
