Calibre does not have an automatic sync function of the sort you mention. However there are a few (relatively new) features that might get you a long way towards what you want.

- The Book Sync GUI plugin that allow you to easily queue books for later automated uploading to the reader device when it is next plugged in.

- The introduction of a last Modifed field for books so you can see when you last made a change. Sorting on this field can help identify books that might need resending to the device. To make this visible in the GUI, right-click a column header and then Show Column->>Modified
