Nope - I've changed nothing. Totally don't understand why it stopped.
I've checked the network settings - they are fine. If they were changed or a firewall somehow was turned on, VLC Streamer and Briefcase wouldn't work either, but they work fine.
Additionally, I just tried logging into my computer with a new account and re-starting Calibre. Still doesn't work.
