The Sony Reader store has some free comics/manga on the site. You could download a few of those to see how they work out on the reader.
Not sure where to buy comics from other than that.
I know Dark Horse Comics has a thing on their website about a Digital Comic book store coming soon, but I'm going to take a wild guess that it is going to be an app you download onto a device (ipad/smartphone/tablet) and read the comics through that instead of being a downloadable pdf file that could be read on a reader.
