My suggestion:

Try it at home and follow your reading rituals as usual on different lights and conditions, even at bed. Some people say that altering the brightness help a bit, but on my particular case and several other friends, I cannot read for too much time without start having eye strain, even with my Galaxy Tab although is not as noticeable.

Rooting will not fix your eye strain issue but give you access to Android Market where you probably will find Android e readers which give you several background options or night reading mode (black background and white letters)

I use an eink device as a dedicated e-reader instead. It is the closest way to get a pbook experience.
