Maybe you only have an existing EPUB format for that book to convert FROM.

If you have multiple formats available, the the list gives you the option of overriding the preferred (possible best) source.
Note: once you have converted a book, Calibre defaults the conversion to the last used (and used options). There is a reset button near the button, that can be used to set it to use the 'Current' Calibre defaults' (which you may have adjusted to taste.
