One thing that can create outrageously long page turns (10 seconds or more) is a poorly made epub file. Long books definitely require splitting into chapters. I reformat ALL my epubs using Sigil, and while it is fairly rare, I do occasionally run into epubs with all the text contained in a single xhtml file. It does not sound like you have run into anything this bad, so I'm mostly posting this for others who may have.
