This is one of the things that I think is great about the Kindle, compared to the Nook. It is very easy on the kindle. All you do is press up or down on the 5-way directional to make the cursor appear on the screen (press up to make the cursor appear on the bottom if the word is closer to the bottom, press down to make the cursor appear at the top of the page if the word is closer to the top). Then use the direction pad to place the cursor in front of the word and the definition pops up automatically. It only shows a few lines of the definition. If you want more, press enter to see the full dictionary entry, then the back button to return.
So, if you count holding down the down button to make it jump multiple lines as "one click", then you need just one click if it's the first word on a line, or two clicks (down, then right) to get to any other word. Plus one click (the back button) to make it go away.
