Turning it off forces it to boot up, which takes a lot of power as the processor is taxed fully, just waking up from sleep is like turning a page
Turning it off "might" be usefull if you're not going to touch it for a long time (read: weeks) although I've had mine on sleep mode for a week or two and it was still pretty fully charged.

Not on the Kindle 3, it doesn't.

the sort of "off" you get with a 7 second hold is barely distinguishable from sleep.
The only thing I notice is the lack of a screen saver.
Y'all might want to try the 7 second hold to see what it actually does. It's nowhere near the same as turning off a computer.
It wakes up the same as from screensaver mode.
