How about better sealing between the screen and the bezel? Mine is constantly getting little pieces of hair wedged in there and it is a pain to remove them.

Maybe some kind of built in latch mechanism on the back to allow more secure attachment to a case (or other attachments such as a strap).

Small trackpad or rollerball instead of the 5-way rocker for better cursor movement)

Louder speakers. I use the auto-read feature in the car and it isn't nearly loud enough. I have to hold it next to my head. Either that or add bluetooth support so you can use wireless headphones.

The printing on the keys needs to be molded through the keys rather than just silk screened on the top. I imagine Amazon is taking A TON of Kindles back due to rubbed off printing.

Allow us to turn 3g off and leave wifi on for better battery life.

I agree with Desert Grandma that a strap on the back would be nice, but I think it should be best left up to aftermarket accessories. My attachment idea on the back would be something that a strap accessory could attach to.

A gmail plug in app would be great. No one wants to go through their browser to check their email. A dedicated app would be much better and could be tailored to the specific strengths and weakness of the Kindle.

A RSS feed reader app that would sync your feeds automatically.

This, I think, would be the best idea...

How about establishing a Kindle 3 Web Page Standard so that web designers can say that their page is "Kindle web browser compliant". Kindle could verify this and then have a web portal that has links to all kindle friendly web pages. Of course they probably won't do this because they don't want to increase that "free" 3g usage.
