The screensaver hack, with a bit of extra work, will let you do it.
Go to Google Images and search for your book cover -- e.g., "Dune Book Cover"
Scroll through the resulting images and save (right click, usually) the one that is most detailed/has the highest pixel count
Open it in a photo editor and resize to 600 x 800. Save as a .JPG. You can do this for free in Picasa 3.8, which now comes with Picnik as an editor.
Move it to the screensaver folder of your Kindle. Delete (or move to another "pending" folder, so you can restore them later) all the other images in this folder, so that, each time you put your Kindle to sleep, this book jacket shows up as the screensaver.
Restart (NOT update) your Kindle.
If you're reading more than one book at a time, repeat all of the above (but you will have to wake and put your Kindle to sleep a number of times to cycle through the book cover images if you care about getting the right one).
