Yes, you do. Calibre keeps a copy of the cover separate from the books, because some book formats cannot store covers (e.g., txt). If you had set the cover before the conversion to epub, then the cover is in the epub.
An epub is really a zip, and contains a lot of stuff. You might try getting an epub editor like sigil to see what is in one. Or perhaps the tool mentioned in this post
