I've tried iTunes 10 with calibre 0.7.17 under both OSX and Windows, and it works for me in both environments.
Be aware that when launching iTunes 10 for the first time, your database will be updated, which can take quite some time, depending on the size of your library. This happens before calibre can connect.
Try this - launch iTunes first, and let it do whatever updating it needs to do. When that's complete, launch calibre, then try connecting to iTunes.
If it still fails, file a ticket.
