Calibre & Kindle

I had my Kindle for some time before I installed calibre on my Mac and moved all my library management over to calibre. After a steep learning curve I now have everything setup exactly as I want but do have a few questions:

What criteria does calibre use for determining if a book on the Kindle is the same as one stored in the calibre library?

Does calibre synchronise changes made to metadata of books in the calibre library to the book on the Kindle when it is connected via USB?
